Activation of NFkB by different stimuli, including TNFa and PMA, requires the dissociation of the inhibitor protein. Dissociation is regulated by the phosphorylation of IkB on serine residues 32 and 36 which targets IkB for ubiquinitation and proteasome mediated degradation. The NFkB complex then translocates to the nucleus and activates transcription of responsive genes.
